HPLC法测定脑脊液中去甲万古霉素的浓度

摘要: 目的 建立HPLC法测定人脑脊液中去甲万古霉素浓度。方法 采用ODS hypersil C18柱,甲醇0.05mol/L-KH2PO4(16:84,pH=5.0)为流动相,检测波长280nm,以替硝唑为内标对去甲万古霉素进行定量测定。结果 脑脊液中去甲万古霉素在1～60mg/ml范围内浓度与峰面积具良好线性(r=0.9993),平均回收率为98.70%±2.65%,日内及日间RSD分别为1.73%～1.95%和3.62～6.78%。Abstract: OBJECTIVE To establish a HPLC method for the determination of norvancomycin in cerebrospinal fluid.METHODS With tinidazole as the internal standard, the content of norvancomycin was assayed on a ODS-hypersil C18 column with a mobile phase composed of methanol-phosphate buffer solution(16:84,pH=5.0). The detection wavelength was 280nm.RESULTS Concentration compared with the peak area ratio, the linearity was obtained over the range of 1～60 μg.ml-1 (r=0.9993).The average recovery was 98.70%±2.65% and the intra-day and the inter-day RSD were within 1.73%～1.95% and 3.62～6.78% respectively.
