原花青素对H2O2致PC12细胞氧化损伤作用的保护研究

摘要: 目的:探讨原花青素对H2O2致PC12细胞氧化损伤的保护作用。方法:使用H2O2造成PC12细胞损伤,MTT法测细胞活性,流式细胞仪检测细胞凋亡率,免疫组化法检测细胞中Bax与Bcl-2的蛋白表达。结果:原花青素能提高H2O2损伤后细胞的活性;能减少细胞凋亡;能够增加Bcl-2蛋白表达,减少Bax蛋白表达。结论:原花青素对H2O2致细胞损伤具有保护作用,能够减少细胞凋亡,其机制可能与其增加Bcl-2蛋白表达,减少Bax蛋白表达有关。Abstract: Objective :To investigate the protective effect of procyanidine on the injury of PC12 cells induced by H2O2. Methods :The PC12 cells were injuried induced by H2O2.The cel1 activity was determined by MTT.The cell apoptosis rate were detected by flow cytometry.Proteins of Bcl-2 and Bax were detected by immunohistochemistry. Results :Procyanidine increased the survival rate and decreased the apoptotic rate of PC12 induced by H2O2.Procyanidine increased Bcl-2 expression and decreased of Bax expression in PC12 induced by H2O2. Conclusion :Procyanidine can inhibit the apoptosis of PC12 induced by H2O2,which might be correlated with increasing Bcl-2 expression and decreasing Bax expression.
