HPLC-TOFMS法快速分离与鉴别丹香冠心注射液中的化学成分

摘要: 目的 采用HPLC-TOFMS对丹香冠心注射液中化学成分进行快速分离与鉴别。 方法 色谱柱Agilent Zorbax SB-C18柱(3.0 mm × 100 mm, 3.5 μm),以0.5 %醋酸(A)-乙腈(B)为流动相,梯度洗脱(v/v),0~25 min, 2%~33% B; 25~30 min, 33%~95% B,进样量5 μl,流速0.5 ml/min,柱温为30 ℃,飞行时间质谱配有ESI离子源,质量数扫描范围m/z 100~1 000。 结果 采用HPLC-TOFMS方法对丹香冠心注射液制液进行了在线化学成分分离,共鉴别出丹香冠心注射液中31个化学成分。 结论 采用HPLC-TOFMS方法对丹香冠心注射液的化学成分进行快速分析鉴别,可为中药丹香冠心注射液的药效物质基础和质量控制研究奠定基础。

Abstract: Objective To separate and identify the components in Danxiangguanxin injection by high performance liquid chromatography time-of-flight mass spectrometry (HPLC-TOFMS). Methods Agilent Zorbax SB-Cl8 column (3.0 mm×100 mm, 3.5 μm) was used for separation and identification of chemical components in Danxiangguanxin injection with a mobile phase of 0.5 % acetic acid (A) and acetonitrile (B) in gradient elution. 0~25 min, 2%~33% B; 25~30 min, 33%~95% B; the flowing rate was 0.5 ml/min and the injection volume was 5 μl. And the time-of-flight mass spectrometer was equipped with an EIS ion source. Scanning mass range was ranged between m/z 100~1000. Results Thirty-one chemical compounds in Danxiangguanxin injection were identified unequivocally. Conclusion Chromatographic demonstration of chemical compounds in Danxiangguanxin injection could provide foundation for the further metabolism and action mechanism study of Danxiangguanxin injection.-
