Determination of glutathione and oxidized glutathione by HPLC

  • Objective To establish a method to determine GSH and GSSG by HPLC. Method The analytical column was Hypersil C18. The mobile phase consisted of 25m mol/L KH2PO4 (pH2.5):tetrahydrofuran (100:1). The flow rate was 1.0 ml/min. Column temperature was 30 ℃ and the detective wavelength was 230 nm. Result The calibration curve of GSH was linear within the range of 3.12 μg/ml~2490.0 μg/ml (r=0.9999) and the average recovery was 101.84%. The calibration curve of GSSG was linear within the range of 3.25 μg/ml~975.0 μg/ml (r=0.9999) and the average recovery was 97,84%. Conclusion The method was proved to be reliable and accurate,which could be used to determinate GSH and GSSG simultaneously.
